AIM: Simultaneous estimation of multi component containing formulation by UV spectrophotometry.

  • Requirements:
    Instrument: - Shimadzu UV 1700 UV-Visible spectrophotometer,
    (LCGC) Electronic Balance
    Apparatus: - 10ml, 100ml, volumetric flask, funnel, pipette, stirrer, measuring cylinder, etc.
    Chemicals: - Methanol and distilled water.
    Sample: -Bulk drug Ibuprofen and Curcumin
    Principle: - Simultaneous equation method: -
    Ibuprofen and curcumin where analysed simultaneously using distilled water as a solvent. The λmax.Of
    Was found be ____nm and ___nm respectively.
    Procedure: -
    Preparation of standard stock solution:- Weigh accurately about 10 mg of reference standard Curcumin and Ibuprofen and transfer to 100 ml of volumetric flask and dissolve in 20 ml methanol and further volume make up to 100 ml distilled water to give stock solution of concentration 100 µg/ml Ibuprofen and Curcumin respectively.
    Selection of analytical wavelength: - Prepare solutions of 10 µg/ml Ibuprofen and 10 µg/ml Curcumin separately. These solutions are scanned in spectrum mode of instrument from 400nm to 200 nm. The maximum absorbance of and observed at ___ nm and ____ nm respectively.
    Preparation of calibration curve: - Make the further dilutions by pipetting out 0.2, 0.4, 0.6, 0.8, 1.0 ml of the standard stock solution of Ibuprofen in 10 ml volumetric flask and make up the volume up to 10 ml with distilled water to give concentrations 2, 4, 6, 8, and 10 µg/ml. of Ibuprofen. Also pipetting out 1, 2, 3, 4, 5 ml of the standard stock solution of Curcumin in 10 ml volumetric flask and make up the volume up to 10 ml with distilled water to give concentrations 10, 20, 30, 40, and 50 µg/ml. scan all the dilutions in spectrum mode of instrument from 400 nm-200 nm. Measure the absorbance of each solution at respective theirλmax. And prepare the calibration curve for both the drugs. The linearity of Tramadol and Paracetamol is found to be 2-20 µg/ml and 10-50 µg/ml. Determine the absorptivity values of Tramadol and Paracetamol at λmax.of both the drugs.
    Determination of concentration of standard mixture:-The mixed standard solutions in Beer Lambert’s range for each drug in the ratio of 1:5 ofIbuprofen and Curcuminare prepared. The mixed standard solution having concentration 2-10 µg/ml Ibuprofen and 10-50 µg/ml Curcumin. The mixed standards are scanned in spectrum mode of instrument from 400 nm to 200 nm. The absorbance of solutions are measured at ____nm and ____nm. The concentration of each drug present in mixed standard solutions are calculated by using simultaneous equation method.
    Result:
    The concentration of and in given mixture by using simultaneous equation method was found to be _______ µg/ml and _______ µg/ml respectively.
