AIM: To determine the unknown concentration by fluorimetry.
Requirements:
Instrument: - Photofluorometer, Electronic balance (LCGC).
Apparatus: -Conical flask, beaker, stirrer, funnel, R.B.F., Pipette.
Chemicals: - Sulphuric Acid (0.1M).
Sample: - Marketed Quinine Sulphate tablet.

  • Principle:
    Fluorescence is the emission of light from a molecule, which return to its normal ground state from lowest vibrational level that is V0 of the excited singlet state. Excitation of molecule is achieved by exposing them to a light source of specified wavelength. Intensity of fluorescence is directly proportional to the concentration of fluorescent species. The relationship is observed for dilute solution only.
    Procedure:-
  • Weight 10 mg of Quinine Sulphate.
  • Dissolve it in 100 ml 0.1 M Sulphuric Acid.
  • Remove 1ml from it and dilute it to 100ml with 0.1M Sulphuric acid solution. This will give 1µg/ml solution of Quinine Sulphate.
  • Pipette out 1ml of the resulting solution and dilute it to 10ml to give the concentration of 100µg/ml like this prepare the dilutions of 200 µg /ml , 300 /µg ml , 400 µg /ml , 500µg /ml , 600µg /ml of Quinine Sulphate.
  • Measure the fluorescence of each solution and unknown, adjusting zero reading with blank solution and adjust 100 reading with 600 µg /ml as a standard reading.
  • Formula:
    U-B=Conc. Of unknown
    S-B=Conc. Of standard
    Conc. Of unknown=U-B X conc. Of std S-B
  • Find the concentration of unknown sample using formula.
  • Preparation of 0.1M Sulphuric acid: - Add 5.4ml of conc. Sulphuric acid slowly in 1000ml volumetric flash containing water and dilute up to mark.
    Result: Concentration of unknown sample by calculation using formula was found to be______________ µg/100ml.
